The Bio-Rad QX200 ddPCR system combines water-in-oil emulsion droplet technology with microfluidics technology. The QX200 system consists of two parts-a droplet generator and a droplet analyzer. The droplet generator can generate 20,000 uniform nano-upgraded droplets for each sample. The target fragment and background sequence are randomly distributed in the droplets. Each droplet is an independent reactor. The droplets were then transferred to a 96-well PCR plate and amplified on an ordinary PCR machine. After PCR amplification, the droplet analyzer performs fluorescence detection on the droplets one by one. The fluorescence signal intensity of a positive droplet containing at least one target DNA or RNA molecule is increased compared to a negative droplet. Finally, according to the Poisson distribution, the absolute copy number of the target molecule is given by the ratio of the positive droplets. The QX200 system is compatible with TaqMan hydrolysis probes and EvaGreen detection. In addition, the flexible design allows users to combine ultra-sensitive nucleic acid detection by combining wells or run one sample through one well to achieve high-throughput sample detection.
